Tiole and Saint Trina are NPCs from the Shadow of the Erdtree add-on in Elden Ring, whose side quests are closely related to each other. Tiole is one of the Extinguished, a master of poison making, summoned to the Shadow Realm by Mikella the Unclouded. Tiole used to worship Saint Trina until Mikella hid her behind his seal, causing him to forget about her.

How to start the Tiolier quest in Shadow of the Erdtree
The quests of Thiolier and Saint Trina can be completed if you have not yet visited Enir-Ilim.
Where to find Thiolier
To start Tiole's quest, you need to find him at the grace “Cross by the road with columns”. You can find this point by moving from the “Cross by the three roads” point to the northwest across the bridge over the Ellak River until you reach the “Front side of the castle” teleport.
From there, first head southeast until you reach a fork, take the left path and go forward until you see the “Cross by the Columned Road” grace and Tiole sitting in the distance.
Talk to him. He will tell you that he deals with poisons, and then you will have the opportunity to trade with him.
To start the quest, as well as get Tiole's marker on the map, you need to deliver him the Black Molasses, picking it up from the merchant Moore. Moore's quest and all the details on delivering the Black Molasses can be read in this article.
After receiving the molasses, Tiole will reward you with Tiole's Potion, which can be useful in the Priestess of the Dragon Communion quest if you want to get its alternate ending.
Tiole's storyline will not advance further until you reach the “Shadow Haven” location, where you will have to fight another boss.
Upon reaching the “Shadow Haven, Main Gate” point, the following inscription will appear: “Somewhere a great rune has split… And powerful spells have dissipated“.
After this event, return back to Thiolier, who will be waiting for you in the same place.
With Mikella's spell broken, Tiole begins to remember Saint Trina and worry that Mikella will punish him for loving her.
Conversation with Leda the Needler
If you also talk to Leda at this time, who is at the grace of the “Cross by the Road”, she will also notice that Mikella's spell has dissipated. Leda will decide that it is necessary to find and destroy all those unworthy of Mikella.
Since we learned from Tiole himself about his love for Saint Trina, we can suggest that Leda kill him. This choice will not affect Tiole and Trina's quest in any way. Leda will say that although Tiole's love for Michelle is weak, he is not capable of betrayal, since Saint Trina also loves Michelle.
Talk to Tiole again, who will tell you that he is heading south. Mur told him that dark purple water lilies have bloomed on the southern coast, which means that Saint Trina may be located there.
How find Saint Trina
Find the Purple Garden
The Holy Trine is located in the hidden location of the Purple Garden, which is located on the southern coast. How to find the coast map, if you have not yet opened it, is described in detail in this article.
Reach the grace point “Azure Coast” and go south, passing the Death Dragon. When you reach the cliff, turn right and follow the road with glowing worms until you see a giant. Go forward, keeping to the right until you notice a huge rift.
Descend the rocks into the rift, reaching the “Rift” point. Go forward into the cave, exit into a huge dungeon and find a new save point “Rift in a stone sarcophagus”.
Follow southeast until you reach a cliff that should collapse under your feet.
Moving northeast you will find the next point “Cross in the Rift”. Go down and also follow northeast, beware of stone birds and worms. Keep north, reaching the next save point “Rest in the Rift”.
After touching the grace, go further, the road will be linear, and you are unlikely to get lost.
On the way, you will be met by an enemy, the Bastard Warrior, after defeating whom you will receive a new prayer, “Layered Rings of Light”, which inflicts continuous damage to enemies.
Going east you will see another point “Deep Rift”, around which animals will sleep. Go up the huge stairs, and soon you will reach the spirit and the sculpture of a huge horned head. Do not be afraid and jump down from it.
This will take you to the Violet Garden. You need to fight the next boss, the Rotting Knight. There will be a summon sign for Tiole to help you in the fight, but only if you talked to him before you went looking for Saint Trina.
After the knight is destroyed, the Violet Garden teleporter will open. Go further into the small cave and you will see Saint Trina. If you do not find Tiole there, while away the time at the grace and return back. Tiole will be lying not far from her, immersed in sleep.
Hear the messages of Saint Trina
Approach Trina. It is impossible to talk to her and when interacting with her there will be only one option – “Drink the nectar”. After taking the nectar, your character will instantly die and respawn at the previous save point. Go back to the cave, collecting lost runes along the way. Tiolier will warn you not to drink the nectar, as he will believe that only he can withstand it.
Continue drinking the nectar until you hear Trina's first message asking you to stop Mikella.
Talk to Tiole and tell him Trina's words, which he will not believe. Repeat the words again and Tiole will become even more angry.
Continue drinking the nectar until you hear another message from Trina warning Mikella against becoming a god.
After resurrecting once again, you will suddenly hear the voice of Tiolier, who will decide to attack you. It will be easy to deal with him, since he is really weak, despite the fact that he will use poisons in battle.
After defeating Tiolier, the inscription “Tiolier died” will appear on the screen. You will also receive an item – Smile of Saint Trina. This talisman increases the attack power near sleeping enemies. But this is not the end of the quest.
Return back to the cave where you need to talk to the dying Tiole. He will ask you to convey Trina's message to him again, but you will not be able to do this now.
Drink the nectar again and listen to the saint's last words asking you to kill Mikella. Tell Tiole about this and leave the Violet Garden.
Quest finale
Deal with Leda and her alliesÂ
In order to complete the quest of Saint Trina and Thiolier, open the passage to Enir-Ilim and proceed to the travel point “Antechamber of the Chamber of Purification”. Needlewoman Leda will be waiting for you there and will challenge you. Before the battle, you can call Tiole and Ansbach to help you in turn.
After defeating Leda and her allies, move on to a very strong boss – Future Consort Radan. Before entering the fog, you will again see a sign with which you can summon Tiole. Enter the arena and try to win.
If you can't find Tiole in the arena after the fight, while away the time at the grace and you'll find him lying dead. You can loot the following items from him: Tiole's Hidden Needle, as well as his mask, robe, gloves, and pants.
This needle has a special skill, “Sleep Forever,” which can put enemies to sleep forever.
Return to the Purple Garden to Saint Trina, who you will also find dead. Take the Flower of Saint Trina, which slightly increases the maximum OK. This will complete the quest of Saint Trina and Tiole.
